Hurricane Sandy and the Politics of Everything - NYTimes.com

In The Conversation, David Brooks and Gail Collins talk between columns every Wednesday.

David Brooks:? Gail, how did you survive the storm? We here in D.C. got off light. I spent much of the weekend stocking up on water, batteries and spicy gum drops in preparation for the worst, but my family had nothing worse than a few hours of lost power.

Gail Collins: I?m stranded in Ohio. Fond as I am of this part of the country, I?m really anxious to get back to my city.

And out of respect for the trauma of the storm I will try to refrain from reminding you that Mitt Romney is on record wanting to get rid of the Federal Emergency Management Agency.

David: I?m not sure that?s quite fair on FEMA. If you go back and rewatch the debate you?ll see that Romney is dodging the FEMA question by going broad and talking about deficits.? John King, the questioner, understood the dodge and tried unsuccessfully to bring him back to the question.? Subsequent commenters seem less aware than King, for whatever reason.

I must say this was the first news event where I thought Twitter clearly outperformed television news. I spent a little bit of time watching TV reporters on windswept beaches clinging to lamp posts. When that got old after five minutes, I followed the storm on Twitter as it swept through lower Manhattan and Brooklyn, possibly the most Twitter-heavy areas on earth. There were reports and pictures from everywhere all at once. It was actually quite moving as the images came in?from the building that lost its fa?ade, videos of transformers exploding, pictures of the Hoboken train station.

The emotional shock was contagious, as people all across the city began to register the power of the storm. And it was like being part of a depleted platoon mid-battle. Every few minutes somebody else on my Twitter field would announce they?d lost power, and they?d vanish.

Gail: O.K., I admit it. I don?t do Twitter. But this may convert me.

David: The best comment I?ve seen so far was on Walter Russell Mead?s consistently excellent Via Meadia blog. In an essay called ?Nature and Nature?s God,? Mead reminds us that New York is usually a place where people are scarcely aware of nature, except that it?s harder to get a cab in the rain. People are busy leading the lives they?ve created for themselves.

But then something like this happens to remind you that it is an illusion to believe that you are in control of your own life. Much larger forces are out there, capable of sweeping you and everything away. ?Somewhere in the future, each of us has an inescapable appointment with irresistible force,? Mead writes. It could be a natural disaster, an illness, or something else, but it?s coming.

    ?

Gail: Wait a minute. I agree that people in New York do not spend a lot of time in direct communication with nature. But I don?t think they fool themselves about the degree to which they?re in control of their own lives. We?re totally aware that disaster could strike at any minute, and that it?s no respecter of rank or wealth. Terrorists attack, criminals attack, runaway cabs attack. I used to work for the tabs so I could go on and on. But it is true that we?re more likely to imagine falling through a subway grate than being carried off by the tide.

David: ?I actually think people in less affluent times built their lives around this awareness. They saved more. Resisted debt more. Raised their kids more strictly because there was less of a cushion if you messed up.

If this storm has any cultural impact ? and they usually only do in the most ephemeral sense ? it will be to remind people to build low and sturdy. I wouldn?t say our government is built to withstand some future shock (too much debt, too much disunity). This election campaign suggests we?re not going to fix that soon.

Gail: You know, we get along so well I sometimes forget we really do have different political worldviews. The message I take away from the storm is that in natural disasters, people can?t help themselves. They need a strong government to swoop in with a rescue operation. A government with a sturdy FEMA that has not been dismantled by a feckless White House of the future.

David: We?re finally less than a week away from Election Day. It?s been a miserable year. I don?t know about you, but I can?t wait for it to be over. Among other things, I?m just sick of the mood that envelops people at the height of campaign season.

Gail: I have to admit I?m a little tired of writing about the campaign, but I find it sad to realize that after next week I may never again have an opportunity to point out that Mitt Romney once drove to Canada with the family dog strapped to the car roof.

David: I have no confidence in my ability to predict what?s going to happen, but I guess I think Obama will be re-elected with somewhere around 290 electoral votes. His leads in places like Ohio are impressively stable. I could be completely wrong though, since Republicans do seem more enthused. But either way, I don?t expect the election will settle any of the country?s divisions and I do look forward to a time when we can talk about other things.

As Sandy reminds us, there are many other things in most people?s lives that are much more important than politics.

Gail: Politics is about everything! How we organize our society. What responsibilities we have to our fellow citizens. I wish it could hold back the tide, but at least politics lets us figure out how we?ll prepare for the cleanup.

    New tick disease in Switzerland

Until now, we knew that ticks primarily transmit two pathogens to humans in Switzerland: the bacterium Borrelia burgdorferi which causes borreliosis and the early-summer-meningoencephalitis virus, which can cause cerebral inflammation. Now, microbiologists from the University of Zurich confirm the existence of another tick disease in Switzerland neoehrlichiosis.

The pathogenic bacteria Candidatus Neoehrlichia mikurensis was first discovered in ticks and rodents in Europe and Asia in 1999. In 2010, Head of Molecular Diagnostics at the Institute of Medical Microbiology Guido Bloemberg and colleagues from Sweden and Germany diagnosed the world's first infections in humans and dubbed the disease "neoehrlichiosis", by detection of the bacterium in the patients blood. Two more cases were identified at the University of Zurich's Institute of Medical Microbiology in October 2011 and January 2012. So far, a total of eight patients have been described in Europe, three of whom come from the Zurich area. They suffered from relapsing fevers of up to 40 degrees, weight loss and general malaise.

    Greater Zurich region risk area

Bloemberg's team studied around 2,000 ticks from the neighborhood of the three Swiss patients, who often spent time in forests and fields. The result: A large number of ticks in the Zurich area five to ten percent carry Candidatus Neoehrlichia mikurensis. "Our study shows that the greater Zurich region is a risk area for neoehrlichiosis, especially for immunocompromised people," explains Florian Maurer from the University of Zurich's Institute of Medical Microbiology. Almost all the patients in Europe to date have had a weakened immune system. However, people with a healthy immune system recently contracted neoehrlichiosis from a tick bite in China, too.

    New test detects infection quickly

"Because the bacteria that cause neoehrlichiosis couldn't be bred in the lab until now and thus no rapid tests were available, many infections might have remained undetected," says Guido Bloemberg. However, help is now at hand with the new DNA test the researchers have developed: It can detect an infection definitively within one working day and also be used for larger test series.

The Swiss patients could be cured fully with a course of antibiotics. Within only a few weeks of beginning the treatment, the microorganism responsible could no longer be detected in their blood. "How well the bacterium is transmitted to humans via a bite from an infected tick, however, still needs to be researched," concludes Bloemberg.

European 'Borgia' drama proves a worldwide hit

SERMONETA, Italy (AP) ? Who doesn't love a wealthy family's bloody saga?

The Italian economy may be struggling but the pan-European television series "Borgia," which is now filming its second season in Italy, is a thriving hit on the world market.

Spanning the late Middle Ages to the early Renaissance period, the show follows the famous Borgia family's rise to power and subsequent domination of the Vatican and southern Europe's political landscape. A winning combination of sex, violence, faith, lust and betrayal, the primarily French-German production has been sold in 85 countries worldwide.

The first season follows the brutally ambitious Catalan Cardinal Rodrigo Borgia (played by U.S. actor John Doman) and his ruthless climb to become Pope Alexander VI. ?The second season, currently shooting, begins in 1494 and its focus slowly moves to his charismatic son Cesare Borgia (played by Irish actor Mark Ryder), the famed warrior prince who inspired Niccolo Machiavelli's "The Prince."

Ryder said the biggest challenge has been taking his character from a whiny young boy in the first season to a "magnificent man of the Renaissance" in the second.

An Associated Press TV crew visited the set near Rome, where there are twenty-four different nationalities among the "Borgia" crew. Doman, best known for his role in the U.S. hit HBO TV series "The Wire" says it's a unique experience to work on such a multi-cultural production.

"We have all come from different places and we are all together working here on this project about a family and we've really kind of gelled as a family," he told the AP.

On the set, a stocky Italian charges across the courtyard of the Caetani Castle and yells, "Action, silenzio!" prompting a tall, blond German to lift a movie camera off a metal frame and an Italian stunt woman to hoist herself onto a horse. Dozens of extras immediately snap to attention.?

Nearby in a black tent, a British director was following the action closely on a monitor while an Irish actor enjoyed a cigarette on his break, careful not to dirty his red velvet cardinal's robes.?

The executive producer, American Tom Fontana, says producers searched far and wide to assemble the talent working on "Borgia."

"We were always determined to have the best actors, the best designers, the most talented people here, regardless of what country they come from," he said. "Everyone who is here knows they are here because we think they are the best."

While the talent comes from around the globe, Irish actor Diarmuid Noyes (who plays Cardinal Alessandro Farnese) says?the work ethic comes straight from their team of directors.

"Every day for eight months (they) are on the ball and they just bash it out, and they are shooting, shooting, shooting, shooting," he said. "They never let up!"

With a budget of ?30 million ($38.7 million), the second season of Beta Film's "Borgia" is being shot over eight months in both Italy and the Czech Republic. Italy alone will feature 20 separate locations.

According to Beta Film, "Borgia" was the highest-rated original series on Canal+ in France and in Italy on Sky Italia. On Germany's public broadcaster ZDF, the company said it was the fictional primetime winner every night.

Romo: We stunk? |?? Coughlin: Clock stuck at 0:01

Tracy Porter, Mike Cerullo, Mike Mallory, Tony Oden,AP

Earlier today, I mentioned that the latest court filing from Saints linebacker Jonathan Vilma in the bounty case contains no new facts or legal arguments.

    Man, was I wrong.

Attached to the brief is the email sent by former Saints assistant coach Mike Cerullo that resurrected the NFL?s investigation.? In a November 2, 2011 message to NFL spokesman Greg Aiello, Cerullo states as follows:? ?So I have info on Saints Joe Vitt Lying to your NFL Investigators on Bounties from 2010, along with proof!!!? I was there, in the cover up meetings, with players and Joe, I love the NFL and want to work there again, but I am afraid if I tell thge [sic] truth I will never coach again in NFL, But I was fired for a situation that the Saints encourage.? All I want is a Job back in the NFL as a QC Coach anywhere, so If talking to you jepodizes [sic] that I will have to get back to you, but The Saints are a Dirty Organization.? Contact me.?

The message contains proof of Cerullo?s alleged bias against Vitt.? As alleged by Vilma earlier in this process, Cerullo had vowed revenge against Vitt for firing Cerullo.? Separately, Cerullo?s zeal to return to the NFL calls into question everything he says.? Also, Cerullo?s reference to being fired ?for a situation that the Saints encourage? requires that claim to be explored and compared to the stated and actual reasons for his termination, in order to properly evaluate Cerullo?s overall perspective and mindset.

The email is attached to a new declaration from Vilma?s lawyer, Peter Ginsberg, who explains that the email was produced by the NFL on October 23, 2012.? Ginsberg contends that the NFL completely removed Aiello?s response.

The Cerullo email is marginally relevant to the effort to disqualify former Commissioner Paul Tagliabue as the bounty suspension hearing officer.? But it will be extremely relevant to the overall appeal and litigation process, given that it makes a full cross examination of Cerullo even more critical to a fair disposition of the case.

    Worry - STDs - MedHelp

Welcome to the Forum.??I'll try to help. Most commercial sex workers do not have STDs and even when they do, most unprotected exposures do not lead to infection.??Further, in your case, condom protected sex is safe sex with no appreciable risk for any STD.??Receipt of oral sex is likewise very low risk.??In addition, I should point out to you that there are no STDs which cause symptoms at 24-48 hours after exposure so any symptoms you have experienced to date are not STDs acquired through the exposures that you describe.??

The wrinkling and white mark you have noted are likely to be normal skin structures that you have not noticed in the past, not signs of an STD.??

Please do not take antibiotics ?just in case?.??Many of our clients do this and it typically confuses things.??If you are truly concerned you can go to a local STD clinic for evaluation.??They are good at STD testing and provide confidential services at little or no cost, sometimes on a walk--in basis and sometimes by appointment,??.??To find your local STD Clinic call the local Health Department.

My advice to you is to not worry.??I hope my comments will help you not to worry.??EWH??

Survivors criticize Myanmar gov't over clashes

SITTWE, Myanmar (AP) ? Survivors of ethnic clashes in western Myanmar lashed out at the government Monday for failing to prevent violence between Muslims and Buddhists that has displaced more than 28,000 people over the last week.

The crisis, which first began in June, has raised international concern and posed one of the biggest challenges yet to Myanmar's reformist President Thein Sein, who inherited power from a xenophobic military junta last year.

The latest violence between ethnic Rakhine Buddhists and Rohingya Muslims began Oct. 21 and has left at least 84 people dead and 129 injured, according to the government. Human rights groups believe the true toll could be far higher.

"The authorities are not solving the problem and soldiers are not defending us," said Kyaw Myint, a Muslim man who took refuge at Thechaung camp outside Sittwe. He fled his home in nearby Pauktaw when it was torched Wednesday.

"I feel as though I am in hell," he said. "We have no one to take care of us, no place to go, and now no job to earn a living."

A 37-year-old Rakhine trader named Maung Than Naing, reached by phone in the village of Kyauktaw, also expressed anger over the government's handling of the violence.

"We are helpless because the government is not dealing with the root of the problem," he said. "We no longer want to live with the Muslims."

Maung Than Naing, who also lost his home in an arson attack, blamed the Rohingya for breaking the calm.

"These poor Muslim people who live hand to mouth burned their own homes so that they enjoy the U.N. aid where they are given shelter and free food," he said.

A tense calm has held across the region since Saturday, Rakhine state spokesman Myo Thant said.

Security had been stepped up in the state, with additional police and soldiers deployed, but he declined to give details.

The priority now is to ensure those who lost homes have adequate shelter and food, Myo Thant said.

The long-brewing conflict is rooted in a dispute over the Muslim residents' origin. Although many Rohingya have lived in Myanmar for generations, they are widely denigrated as intruders who came from neighboring Bangladesh to steal scarce land.

The Rohingya also face official discrimination, a policy encouraged by Myanmar's previous military regimes to enlist popular support among other groups. A 1982 law formally excluded them as one of the country's 135 ethnicities, meaning most are denied basic civil rights and are deprived of citizenship.

Human rights groups say racism also plays a role: Many Rohingya, who speak a Bengali dialect and resemble Muslim Bangladeshis, have darker skin and are heavily discriminated against.

Bangladesh, though, also denies them citizenship. The U.N. estimates their population in Myanmar at 800,000.

Tensions have simmered in western Myanmar since clashes first broke out in June after a Rakhine woman was allegedly raped and murdered by three Muslim men.

The June violence displaced 75,000 people ? also mostly Muslims.

U.N. Resident and Humanitarian Coordinator in Myanmar Ashok Nigam said on Monday that the number of displaced was likely to rise because some people who fled affected areas along the coast by boat last week have yet to be counted.

An estimated 27,300 of the 28,000 newly displaced are Muslims, Nigam said, adding that the U.N. figure was based on statistics from local authorities.

The new numbers bring the total number of displaced in Rakhine state since June to at least 103,000.

redskins Devon Walker Tom Cruise ryan reynolds Star Trek: The Original Series Carlton Morgan Freeman Dead

Final 3 set at WSOP, to play for $8.53M crown

Two poker professionals and a college student have topped nearly 6,600 competitors at the World Series of Poker and are one card session away from poker's richest prize ? an $8.53 million crown.

Arizona State senior Jake Balsiger eliminated Russell Thomas in fourth place early Tuesday in Las Vegas with an ace-king, treading past five meaningless community cards to bump his guaranteed payday up nearly $1 million and setting up the Tuesday night showdown.

His opponents in the no-limit Texas Hold 'em tournament: 24-year-old poker pros Greg Merson of Laurel, Md., and Jesse Sylvia of Las Vegas.

"I can't wait to get back up here and play. I wish we could play it out right now," said Sylvia, who started the session of more than 7 hours with the chip lead but was overtaken by Merson.

"We have a long battle tomorrow," he said.

Thomas won $2.85 million for fourth place, then said after being eliminated that he planned to quit his job as an insurance actuary for Aetna to follow his dream of playing cards.

"Made my company a little mad because I quit my job in a newspaper article, and now I'm doing it in a press conference," he said.

Balsiger, Merson and Sylvia have their sights set on higher scores ? guaranteed at least $3.8 million each as the top three finishers.

Merson vaulted to a dominant chip lead thanks to an opponent's startling error, then finished the session with 88.4 million in chips, compared with 62.8 million for Sylvia and 46.9 million for Balsiger.

Merson eliminated Hungarian poker professional Andras Koroknai in sixth place, calling Koroknai's all-in bet with an ace-king and finding Koroknai with king-queen ? a marginal hand for the situation.

Koroknai won $1.64 million for sixth place, while Merson won 84 million in chips, nearly double that of his closest competitor.

The surprising error came more than six hours into a game with plenty of mental maneuvering as players jockeyed to make the right million-dollar decisions.

Merson said he expected it to happen, and told Sylvia during one of the breaks that he expected Koroknai to bluff away all his chips.

"When short-handed play starts, I think that's my game because a lot of players either play too tight, or they over-adjust," he said.

Poker professional Robert Salaburu was ousted in eighth after a river queen gave Sylvia a higher pair. Minutes later, Michael Esposito went out in seventh, his ace-jack failing to pull ahead of Merson's ace-king.

Jeremy Ausmus, 33, of Las Vegas, placed fifth.

"I never had cards the whole day," Esposito said. "At the end of the day, I'm going home a rich person."

Steve Gee, a 57-year-old poker professional from Sacramento, Calif., was eliminated in ninth place less than two hours into play after testing Thomas with a tough wager for the last of his 11.4 million in chips.

Thomas called and showed a pair of queens, better than Gee's pocket eights.

Gee's ouster meant he made no additional money after a break of more than three months. All nine finalists were paid nearly $755,000, the ninth-place prize, after making the final table in July.

"If I knew he had pocket queens, I would have checked," Gee said after being eliminated. "I'm disappointed. I told myself I wasn't going to play scared."

The six professionals and three amateurs that started the final table tested each other from the start, re-raising chips back and forth in moves that would ultimately result in everyone ? besides the winner, perhaps ? gambling their stacks against one another.

On the first hand of play, Gee bluffed Thomas on a pot worth several million chips, though Thomas held just nine high and wouldn't have been able to consider playing the hand to the end.

Esposito, 44, a commodities broker from Seaford, N.Y., was uncertain about the status of his waterfront home as Superstorm Sandy hit the East Coast.

His son, James Esposito, said neighbors told him the streets are flooded, but the family was focused on the card game in the Nevada desert.

"Odds are likely that it's flooded," he said. "I know I saw a picture of the house three doors down ? the streets are totally flooded."

The chip leader heading into the final table was Sylvia, who began the final stretch with 43.9 million in chips, just over 22 percent of the chips in play.

Chips have no real monetary value in tournament poker. Each player at the final table must lose all his chips to lose the tournament, and win all the chips at the table to be crowned champion.

The tournament began in July with 6,598 players and was chopped down to nine through seven sessions spread over 11 days. Play stopped after nearly 67 hours logged at the tables for each player, with minimum bets going up every two hours.

The finalists played Monday night until only three players remained, leaving the top three to settle the title Tuesday night.

Is It A Good Idea To Use IRA To Pay Debts? | Bankrate.com

    Steve BucciDear Debt Adviser,
I was laid off from my job in February. I went on unemployment until my benefits ran out, and I still don't have a job. My husband works, but without my income, we can't pay our bills. To make ends meet, I took $10,000 out of my individual retirement account. But that's not going to last us very long. I'm 53, and I may not be able to get another full-time job, so I'm thinking of dipping into my IRA again. It worries me, though. Do you get hit with penalties when you use your IRA to pay debts?
    -- Debbie

    Dear Debbie,
I'm glad you wrote to me before you took more money out of your IRA. What you need is a new financial plan, and this may or may not include early withdrawals from your IRA. Just because you're younger than the 59 ? age limit doesn't mean you have to pay a penalty on your early withdrawal. I'll explain this later. First, though, let's do some financial replanning.

It seems you've concluded that you may never again earn what you did before you lost your full-time position. And you can't afford to wait to find out. Given that, it's time to match your spending to meet those new expectations.

I suggest that you start by documenting where all your money goes each month. You'll be surprised at how quickly you'll find waste and fluff. No one likes to reduce expenses, but it may be necessary to pay your bills. Additionally, if you are using your credit cards to extend your income, you must stop. Use cash or a debit card so you don't add to your existing debt load.

If you need help with trimming expenses, contact a reputable, nonprofit credit counseling agency at the National Foundation for Credit Counseling or the Association of Independent Consumer Credit Counseling Agencies. A certified credit counselor will review your expenses and income and make suggestions on other budget cuts. A counselor also may help with lowering payments on your credit card debt.

If your bare-bones expenses still outstrip your income, you might consider taking IRA withdrawals to bridge the gap as long as it is a temporary fix. But watch out: If you keep taking money out of your IRA, you'll have a horrible retirement. As part of a short-term bridge, the Internal Revenue Service has rules that allow you to take early withdrawals from your IRA without a penalty.

As a last resort, you might consider a Chapter 13 bankruptcy filing. This could help you keep your house, and your unsecured creditors would have to settle for what the judge believes you can afford to pay over five years. You must be sure, however, that you can afford your current mortgage and all other expenses with your new income level. If not, it might be better to sell your home and downsize your living arrangements.

    Good luck!

Mechanism found for destruction of key allergy-inducing complexes, researchers say

ScienceDaily (Oct. 28, 2012) ? Researchers have learned how a human-made molecule destroys complexes that induce allergic responses -- a discovery that could lead to the development of highly potent, rapidly acting interventions for a host of acute allergic reactions.

The study, published online Oct. 28 in Nature, was led by scientists at the Stanford University School of Medicine and the University of Bern, Switzerland.

The new inhibitor disarms IgE antibodies, pivotal players in acute allergies, by detaching the antibody from its partner in crime, a molecule called FcR. (Other mechanisms lead to slower-developing allergic reactions.)

"It would be an incredible intervention if you could rapidly disconnect IgE antibodies in the midst of an acute allergic response," said Ted Jardetzky, PhD, professor of structural biology and senior investigator for the study. It turns out the inhibitor used by the team does just that.

A myriad of allergens, ranging from ragweed pollen to bee venom to peanuts, can set off IgE antibodies, resulting in allergic reactions within seconds. The new inhibitor destroys the complex that tethers IgE to the cells responsible for the reaction, called mast cells. Severing this connection would be the holy grail of IgE-targeted allergy treatment.

The first time a potential allergen enters the body, some people respond by making allergen-specific IgE antibodies. These antibodies stick around long after the initial allergen is cleared from the body. Most of the antibodies get snagged by IgE-specific receptors called FcRs, which are exposed on the surface of mast cells. The mast cells are then primed to react the next time a person encounters the allergen.

Dissociation of this IgE-FcR interaction is a sought-after goal of allergy treatment for a good reason: IgE-coated mast cells are grenades of histamine, and re-encountering the allergen is equivalent to pulling out the clip. When an allergen makes a return visit, it binds to the pre-loaded IgE on the mast cell surface, triggering the release of inflammatory mediators -- including histamine -- that promote the allergic response. As allergy sufferers are well aware, these nasty reactions can occur within a matter of seconds. In a severe allergic response, sudden anaphylactic shock and death can be the result.

The key to actively disabling the allergic response lies in the separation of IgE from the FcRs on the surface of mast cells. But separating these dangerous couples is a tall order because their interaction is extremely stable -- sensitizing the mast cells for weeks. Currently available treatment using omalizumab (an anti-IgE antibody sold under the trade name Xolair) can block new interactions between IgE and FcR, but it is not designed to pry the molecules apart once they've formed a bond on the surface of a mast cell. So Xolair can dampen the allergic response, but as stated on the product's website: "Xolair is not a rescue medicine and should not be used to treat sudden asthma attacks."

While simply blocking IgE binding is helpful for some allergy sufferers, when it comes to the rapid quenching of an acute allergic response, "what you'd really like to do is get rid of it," said Jardetzky. Along with scientists at the University of Bern, his team discovered that an engineered protein inhibitor called DARPin E2-79 stripped IgE from the mast cell receptor. Using this inhibitor, "an interaction that normally lasts for hours or days in terms of its stability is stripped off in a matter of seconds," said Jardetzky.

DARPin E2-79 is one of a family of engineered inhibitors containing protein-binding regions called ankryin repeats. While Jardetzky's group was using structural biology and biophysical approaches to probe the weak spots in the IgE-FcR interaction, scientists at the University of Bern were tinkering with DARPins that dampened IgE's disastrous effects. The collaboration of the two groups resulted in the characterization of DARPin E2-79, an inhibitor that goes beyond mere blockade to actively disassemble the IgE-FcR power couple.

Jardetzky's group solved E2-79'S structure and used this information to model its interaction with the IgE-FcR pair. Then, using sensitive biochemical techniques that detect step-by-step binding interactions between molecules, the teams were able to tease out the mechanism that the inhibitor uses to break the IgE-FcR bond.

The researchers found that E2-79 hastens the separation of the two molecules by taking advantage of a moment of weakness in the relationship between IgE and FcR. IgE maintains its interaction with FcR using two contact points, and occasionally one of these points releases while the other one keeps the pair together. Normally this brief looseness isn't enough to separate the couple, but E2-79 can swoop into the small space between them, effectively driving the couple apart.

While E2-79 is the first molecule to display these IgE stripping characteristics, Jardetzky hopes that this work will stimulate the discovery of smaller compounds capable of working even more efficiently. Drug developers generally expect large macromolecules like E2-79 to be less potent than small molecule inhibitors and unlikely to be able to disrupt complexes, so the fact that E2-79 worked so well was a surprise. Small molecules are more amenable to oral administration, and are easier and cheaper to manufacture than large macromolecules. "Now we're in the hunt for a small molecule that could have this kind of activity. That would be the real hit," said Jardetzky.

The discovery of E2-79's mechanism of IgE inhibition could lead to rapid discoveries from other labs as well. Now that scientists know what mechanism to look for, they may be inspired to dig back through freezers full of IgE inhibitors that were identified years ago, said Jardetzky. In the light of techniques described in this study, perhaps once-neglected inhibitors will show new promise in the treatment of allergic disease.

The study's primary authorship was shared between Beomkyu Kim, PhD, a Stanford graduate student, and Alexander Eggel, PhD, at the University of Bern. The other Stanford co-author is research assistant Svetlana Tarchevskaya.

The study was funded by the National Institutes of Health, the American Asthma Foundation and the Swiss National Science Foundation.

OUTFLANKED AND SLOW: How E-Ink Got ... - Business Insider

SINGAPORE (Reuters) - Amidst our growing love affair with the tablet, spare a thought for its increasingly shelfbound sibling: the e-reader.

Take Taiwan's E Ink Holdings Inc, which makes most of the monochrome displays for devices such as Amazon.com Inc's Kindle and Barnes & Noble Inc's Nook. After five years of heady growth during which shipments rose 100-fold, it got a jolt at the end of 2011 when monthly revenues dropped 91 percent in two months.

"The bottom fell out of the market," says E Ink Chief Marketing Officer Sriram Peruvemba.

E-readers initially benefited from their reflective displays, which can be read in sunlight and require very little power. But the success of Apple Inc's iPad, improved backlit displays, power-saving technologies and new smaller tablets all point to one thing: the e-reader has become a transitional technology.

Think the harpsichord, replaced by the piano. Or Apple's iPod music player, which helped popularize the MP3 player until the arrival of the iPhone, which could play music but also do a lot of other things.

Now electronic paper companies like E Ink are scrabbling for new ways to sell the technology or in some cases, are pulling the plug entirely.

A recent survey by the Pew Research Center found that of those Americans over 30 who read e-books, less than half do so on an e-reader. For those under 30, the number falls to less than a quarter.

Analysts have cut forecasts, sometimes dramatically. IHS iSuppli predicted last December there would be 43 million e-readers shipped in 2014. When it revised those numbers last month, the estimate was lowered by two thirds.

By contrast, Morgan Stanley in June doubled its estimates for 2013 tablet shipments, predicting 216 million compared with its February 2011 forecast of 102 million.

"Frustratingly for the E Ink guys, it's a transition device," says Robin Birtle, who runs an e-book publishing company in Japan. "Kids won't need this."

    POLE POSITION

Companies giving up the ghost include Japanese tyre maker Bridgestone Corp which ended e-paper production this year after six years in the business, blaming falling prices and the rising popularity of tablets with LCD displays. Its partner Delta Electronics Inc also said it was pulling out.

Qualcomm Inc, which snapped up two startups and launched several devices including the Kyobo Reader in South Korea, told investors in July it would now focus on licensing its Mirasol display technology.

UK-based Plastic Logic said it had stopped making e-readers and was now looking to license its display technology for devices such as credit cards.

That leaves E Ink, which this year bought one of its few remaining competitors, SiPix Technology, in pole position.

Not all the news is bad. A new generation of e-readers with front lighting, which allows reading in the dark, is hitting the market. The Kindle Paperwhite sold out quickly and that device and the basic $69 Kindle e-reader are the No. 2 and No. 3 top selling products on Amazon, based on unit sales. Amazon also recently launched Kindles in two big new markets - India and Japan.

E Ink's revenues have picked up somewhat from late last year and Chief Executive Scott Liu is promising good numbers when the company announces quarterly results on Wednesday.

But E Ink is betting its future, not on consumers buying more e-readers, but elsewhere - including education, an area it sees as essential to growth.

It has started to focus on adding features for classrooms, such as a master device to control which pages students look at, preventing them from flipping ahead to, for example, an answers page. Amazon this month announced a push to get Kindles into U.S. schools, selling e-readers at bulk discount.

But it will be an uphill battle. For one thing, Apple has stolen a march in the United States, saying that 80 percent of the country's "core curricula" is available in its digital bookstore. And while educational institutions are investing in e-books, they're not necessarily investing in e-reading hardware.

In Singapore, for example, one university library has dedicated 95 percent of its budget to e-books. But the country remains one of the few where the Kindle is not available, suggesting that those e-books are not being read on dedicated devices.

E Ink also hopes to see its technology in more devices than e-readers. Over the years E Ink displays have appeared in watches, on a Samsung cellphone keypad and on USB drives. One e-ink sign in Japan survived the 2011 earthquake and tsunami and was able to display emergency contact and route information long after other powered-displays fell dark.

Peruvemba travels the world to trade shows peddling an impressive array of prototypes he hopes to tempt manufacturers with, from a music stand with a built-in e-reader to a traffic light. Says CEO Liu: "I've told our people that in five years non e-reader applications will be as big as the e-reader applications."

This makes sense, analysts say. "We have dialed back our take on them," said Jonathan Melnick of Lux Research. "But we still think the technology is going to have a future. It's just not going to be in e-readers."

    OUTFLANKED AND SLOW

But not all are so optimistic. Not only has E Ink been outflanked by the emergence of the tablet, it's also been slow to innovate.

Although the screens of the latest Kindles refresh faster than earlier models, critics say they still look a little old-fashioned alongside displays from Apple or Samsung Electronics Co Ltd.

"I don't see any significant improvements in the technology in the past few years," says Calvin Shao of Fubon Securities.

E Ink's own history is not encouraging. It took a long time for e-ink to emerge: Xerox had dabbled in it since the 1970s but it was only in the late 1990s that physicist Joseph Jacobson thought of mixing a dark dye and particles of white titanium dioxide in microcapsules. Stimulated by an electrical charge- a process called electrophoresis - one or other would move to the top to form shapes.

Even then it took seven years and $150 million for the company he founded, E Ink, to create its first e-reader, and another two years to tease out production problems for its first customer Sony Corp.

And then it took Amazon's heft to persuade the public to adopt the e-reader by adding a compelling range of books, wireless connectivity and the promise of instant downloads.

E Ink says it is undeterred and intends to play a more central role in any new industry it finds a foothold in. "For our new products we will no longer be a component player," said CEO Liu.

Its chances of success are limited, says Alva Taylor, who uses E Ink as a case study for his classes at the Tuck School of Business at Dartmouth College. "The success rate for companies with a technology searching for a solution is pretty low."

    - Others: heal, cure, buff, change equipment, use item...

    - defend/dodge,

    - Charge,

Attack is to deal damage. If you attack someone, I shall roll a pair of six sided dices and determine the strength and effectiveness of your attack. For example if I roll a 12, then it?s 12 points damage, which is a critical damage

You can be creative with your character style of attack but you need to write the effect of your attack.

Example on how to write your character attacks someone:

A ran around B a quick as the speed of sound to disorient him. Suddenly, A jumped to the sky and kicked the air to propel her down, lunging at B with her spear, attempting to pierce through his helmet and skull which would kill or poison him.

    Or

A quickly moved backwards, attempting to get away from B while shooting at him with her machine guns in both hands. A was aiming for B?s heart, an attack which would kill him instantly if hit directly, if it doesn?t, he will bleed and blood shall be pouring out nonstop.

Noticed I wrote status effect there?

You can only attack one person at a time.

Attack that poison, blind, curse, etcetera, shall be considered a negative status effect attack. To succeed a negative status effect attack, the damage points of an attack needs to be 6 or higher.

Negative status effects shall render the victim?s points to be decreased. Cure shall return the decreased points to normal. Cure and heal don't need dice rolls. They just succeed automatically. Buff actions shall increase the character?s points. Breaking someone?s buff or enchantment shall need to have 6 or more damage points to succeed.

Defend is to weaken a person's attack. Example:

When someone attacks you, I shall role a pair of dice. For instance the dices roll 12. If you don?t defend, the attack shall hit you a direct critical damage. If you decide to use one of your action on defend, I shall roll the pair of dice. For instance the dice roll for defend is 6, then its 12 - 6.

12 for the attack and 6 for defend, equaling it to 6 damage points. From what?s supposed to be a critical damage, it becomes an intermediate damage but negative status effect succeeded because the points are not less than 6. If it was less than 6, the negative status effect would fail.

But if the defend points is bigger than the attack points, there shall be no damage at all.

    Example of Defending or Dodging:

B?s eyes were quick, he jumps back to dodge her attack. (From here you can continue on to your other action for example attacking). B quickly trusted his spear ahead to stab A when she lands, which would injure her.

What happens then, I shall write. The examples I am going to write are:

B didn?t leap quickly enough. A?s spear dug through his helmet, skull, and into his brain. B fell down on the ground with A?s spear still sticking in his head. B had died.

    Or

A?s attack missed and struck her spear on the ground while B?s spear stab through her heart. A died instantly.

You can die without warning. Isn't that fun?

Charge is an action of sacrificing your turn to add + 12 points to the next turn but if anyone attacks you and gets a 6 or above, the charge shall be cancelled.

An attack or charge will be the end of a turn. You cannot charge then attack in 1 turn. You cannot attack then heal in one turn. You cannot attack and cast barriers to add to your defense in one turn. Remember, you only have 2 actions to do.

You can do other actions before attacking or charging.

There will be no time skips or time jumps because on episode 1 of Gods of Elements, there were some time skips that made the story disoriented. If you wish to time skip, please inform me and I shall work something out.

It seems that people prefer to have teleportation and summoning abilities so I?ll allow it but there is going to be a rule for it.

The rules for teleportation and summoning:

Summoning and teleporting is considered controlling the time and space. To control that, you need to manipulate and combine all the elements into one. So in order to use that ability, a lot of energy or a rare item is required.

You can only have your character teleported back to where he had placed a sigil. To place a sigil, someone will have to travel to that place first. You can summon things and people to your side but in a battle, you need to have a success charge to be able to use that ability.

Study finds diabetes raises levels of proteins linked to Alzheimer's features

    Saturday, October 27, 2012

Growing evidence suggests that there may be a link between diabetes and Alzheimer's disease, but the physiological mechanisms by which diabetes impacts brain function and cognition are not fully understood. In a new study published in Aging Cell, researchers at the Salk Institute for Biological Studies show, for the first time, that diabetes enhances the development of aging features that may underlie early pathological events in Alzheimer's.

Specifically, the Salk team found increases in two hallmarks of Alzheimer's-accumulations of amyloid beta (Abeta) and tau protein-in the brains of diabetic mice, especially in cells surrounding blood vessels. Abeta, the misfolded peptide that is thought in part to cause Alzheimer's disease, aggregated inside astrocytes, star-shaped brain cells that, upon interaction with Abeta, release inflammatory molecules that can destroy neurons. Previously, this had not been shown in mouse models of type 1 diabetes (T1D).

"Our study supports and extends the links between diabetes, aging and Alzheimer's," says senior author Pamela Maher, a senior staff scientist in Salk's Laboratory of Cellular Neurobiology. "We show that type 1 diabetes increases vascular-associated amyloid beta buildup in the brain and causes accelerated brain aging."

The findings suggest that the neurovascular system may be a good candidate for new therapeutic targets to treat Alzheimer's in the early stages of the disease.

Alzheimer's and diabetes are two diseases that are increasing at an alarming rate within the U.S. population. Alzheimer's affects one in 10 Americans over 65 years of age and nearly 50 percent of those over 85. Similarly, more than 8 percent of Americans (approximately 26 million people) have diabetes, with the vast majority of those individuals being over 60.

Maher says her team is uncertain of the precise mechanism behind the increase in Abeta and tau in the mouse brain, but their data suggest that changes in astrocytes, as well as other pro-inflammatory processes and the bonding of proteins with sugar molecules (called non-enzymatic glycation), may contribute.

"Astrocytes play a key role in maintaining nerve cells in the brain," says lead study author Antonio Currais, a postdoctoral researcher at Salk. "Both chronic peripheral inflammation and increased non-enzymatic glycation are associated with diabetes, and these changes may act on the brain to alter astrocyte function, which eventually leads to Alzheimer's-like changes."

All nerve cells are closely connected to blood vessels, as they need nutrients---- especially glucose (sugar) and oxygen---- provided by the blood in order to function. Astrocytes facilitate the transfer of nutrients between blood vessels and cells. The buildup of Abeta at sites where astrocytes interact with blood vessels suggest that this could impair the transfer of nutrients. The type of Abeta localization seen in Maher's mouse models is also found in human Alzheimer's patients

To examine the contributions of diabetes to Alzheimer's-related pathology in the aged brain, the Salk researchers induced T1D in two sets of mouse models. One set, known as SAMP8 mice, undergo accelerated aging and develop early deterioration in learning and memory, as well as a number of brain alterations similar to those found in Alzheimer's. The other set, SAMR1 mice, which in this study came from the same gene pool as the SAMP8 mice, age normally.

Using these mice, Maher and her colleagues addressed how T1D interacts with age to contribute to Alzheimer's-related pathology. They showed that T1D elicits a wide range of pathological changes in the brains of both strains of mice, which are exacerbated by premature aging.

The Salk study is the first to show that these modifications are similar to those seen in old nondiabetic SAMP8 mice and to identify unique pathological changes, such as increases in markers for inflammation, in aged, T1D SAMP8 mice. Unlike most mouse studies of Alzheimer's, Maher's mice were not engineered to produce high levels of human Abeta or tau,so all of their observations came from naturally occurringAbeta and tau.
